Driving ASO and siRNA Innovation: Key Success Factors from Discovery to Regulatory Toxicology

This session will examine critical success factors in the development of antisense oligonucleotides (ASOs) and small interfering RNAs (siRNAs), from discovery through regulatory toxicology. Topics include assay methodologies, study design principles, species selection, dosing strategies, and identification of key toxicological endpoints. Regulatory and modality-specific expectations, including NOAEL drivers relevant to safety assessment, will also be addressed.

Bioanalysis of Antibody Drug Conjugates by LC-MS
Speaker: Jordane Biarc, Chromatography, Charles River

Antibody–Drug Conjugates (ADCs) are a promising cancer therapy, with efficacy and safety dependent on controlled payload release. Accurate Drug-to-Antibody Ratio (DAR) measurement and kinetics are essential for understanding ADC behavior. Liquid Chromatography–Mass Spectrometry (LC–MS) offers robust quantification and characterization. Triple-quadrupole instruments monitor specific ADC fragments to calculate DAR over time, while high-resolution MS provides direct visualization of structural changes, delivering insights into ADC integrity and heterogeneity.
