Accugenix® Next Generation Sequencing (Accugenix® NGS)
Accugenix® Next Generation Sequencing for bacterial identification and fungal ID simultaneously sequences millions of individual DNA fragments from a sample and provides a snapshot of the microbiome at that point in time. When investigating mixed species environments, it provides an understanding of microbial populations without the need for any culturing. Additionally, when studying pure isolates and/or sequencing of mixed species environments, NGS provides a deeper resolution of organism genes and variants at a level never before possible.
Investigations needing strain resolution may be opened due to an action level exceedance, a media fill or sterility test failure, or identification of an adverse trend in the EM tracking and trending data. While we maintain an extensive list of species with validated SLST/MLST schemes, if a validated strain typing scheme is not available, a new method may be developed using Sanger sequencing technology. If that development is not possible or information is needed as soon as possible, the Accugenix NGS strain comparison method can be used. This method was developed to compare protein-coding gene sequences inferred from whole genome sequencing data to generate a Strain Typing comparison report, offering the same high-resolution with a greater success rate of comparing target genes.
In addition to strain differentiation, the technology can be used to determine the sequence of entire genomes or targeted regions of DNA or RNA. These data have many applications such as:
- Identifying multiple species in a single sample, reducing the time and cost required for analysis
- When studying mixed species environments, you can understand microbial populations without the need for subculturing
- When studying pure isolates, you can understand organisms’ genes and variants to a degree that is not possible with traditional Sanger sequencing
- Providing identification in cases where Sanger sequencing was unsuccessful

Are there validated methods for Accugenix NGS?
At this time there are very few GMP/validated NGS methods commercially available. When an ID is necessary, Accugenix NGS is better than no ID at all, especially if the original isolate was submitted for sequencing was due to its critical nature.
