In vivo models engrafted with human immune cells are essential for advancing research in immuno-oncology, infectious diseases, and graft rejection. Among these, peripheral blood mononuclear cells (PBMCs) are particularly valuable for humanizing mice due to their rapid engraftment in immunodeficient models.

The NCG-B2m-KO mouse is a next-generation triple-immunodeficient NCG model with a targeted B2m (β2-microglobulin) gene knockout. This modification disrupts β2-microglobulin (B2m) — a key component of the MHC complex — resulting in minimal MHC I expression on cell membranes. As a result, NCG-B2m-KO mice exhibit strong resistance to graft-versus-host disease (GvHD), making them an ideal platform for longer PBMC studies.

By combining NCG-B2m-KO mice with pre-qualified PBMCs, researchers can accelerate their studies with reliable and reproducible results – eliminating the need for additional donor cell qualification and streamlining the path to discovery.
