PBMC Humanized Mouse Models for Preclinical Research

Peripheral blood mononuclear cells (PBMCs) play a crucial role in humanizing immunodeficient mice, offering a powerful platform to evaluate potential therapeutics for diseases such as cancer and infectious diseases. PBMC humanized mouse models are surrogates for human clinical trials, enabling human cellular immune function assessment. These adult immune cells engraft rapidly, making them ideal for short-term studies focused on T cell modulation.

Create Your Own PBMC Humanized Mouse Models with Our Exclusive Kit

Timing is everything in research, but sometimes, you’re not quite ready to begin your study. Our exclusive PBMC Select Humanization Kit offers a convenient, reliable solution for generating PBMC-humanized mouse models exactly when needed.

Our kit gives you greater flexibility in study timing—a crucial advantage when working within a limited therapeutic window, such as slow tumor growth rates. By supplying the mice and cells together, the PBMC Select Kit ensures that humanization aligns seamlessly with your study timeline.

Additionally, our use of pre-qualified and recallable PBMC donors eliminates the need for independent qualification, saving you valuable research time and keeping costs under control.

These donor cells have been screened and pre-qualified for engraftment rate, body weight loss, and study duration. Furthermore, cell numbers are optimized specifically for the NCG mouse model, ensuring consistent and reliable engraftment without a license.

The PBMC Select Humanization Kit is compatible with three NCG Plus Portfolio models to generate these humanized options:

  • HuPBMC NCG
A triple-immunodeficient mouse, lacking functional/mature T, B, and NK cells, with reduced macrophage and dendritic cell function. These animal models can host xenograft cells, tissue, and human immune system components.

  • HuPBMC NCG-B2m-KO
A triple-immunodeficient NCG mouse with a B2m (β2-microglobulin) gene knockout. B2m is a component of the MHC complex essential for MHC I expression. NCG-B2m-KO mice express low levels of MHC I, leading to a delayed onset of graft-versus-host disease (GvHD).

  • HuPBMC NCG-MHC-dKO
A triple-immunodeficient NCG model lacking expression of MHC class I molecule (H2-K and H2-D1) and MHC class II molecule (H2-Ab1). The decreased expression of MHC I and MHC II leads to a significant delay in the onset of GvHD in PBMC humanized mice.
