MALDI-TOF Filamentous Fungi Identification Service: AccuFUN-ID®
Strengthening your EM with Full MALDI-TOF Capability
Identification of filamentous fungi has historically been more challenging than identification of bacterial or yeast isolates due to cell wall composition and complex growth morphologies. Microscopy continues to be one of the most common methods for identification, where technicians mount the fungal specimen, examine the sporulating heads, conidia, and hyphae, and compare to reference pictures. However, this process is very error-prone, time- and labor-intensive, and inconsistent with modern data integrity principles, including ALCOA+ expectations. Relying on microscopic examination can also easily lead to inaccurate identifications during critical investigations of filamentous fungi, increasing risks to patient safety and regulatory compliance.
The MALDI-TOF microbial identification process generates a unique protein spectral fingerprint, which is matched against the GMP-validated Accugenix® MALDI database for identification. AccuFUN-ID® delivers higher accuracy than microscopy or phenotypic methods, faster turnaround, and a cost-effective solution for routine environmental monitoring. By combining our extensive library with robust sample processing using specialized Conidia ID Fungal Plates, we achieve significantly higher identification rates for you.

Specialized ID Fungal Plates for the Cultivation of Filamentous Fungi
While processing bacterial and yeast samples for MALDI-TOF analysis is straightforward with the direct smear or extraction methods, the cell walls of filamentous fungi are more difficult to break up and lead to challenges generating consistently robust spectra. These problems can be overcome using specialized media for the cultivation of filamentous fungi, such as Conidia IDFP (ID fungal plates), followed by the extraction method. Charles River has partnered with Conidia to make these plates available to our global customers.
Get to Deeper Resolution with Fungal Sequencing Services
When species-level accuracy matters, genotypic approaches provide greater precision than phenotypic methods alone. Comparative sequencing of the ribosomal DNA ITS2 region is widely regarded as one of the most accurate and reproducible methods for fungal identification. As a fungal identification service built on stable genetic markers, ITS sequencing delivers consistent, high-confidence results, even for closely related or hard-to-identify fungi.

Frequently Asked Questions (FAQs) About MALDI-TOF for Fungal Identification
-
How does MALDI-TOF for filamentous fungi identification work?
After growing up a sample of filamentous fungi on IDFP agar and extracting the sample, it is spotted onto a MALDI target plate to generate a unique protein spectral fingerprint using mass spectrometry. This fingerprint is compared against the GMP-validated Accugenix® MALDI database to accurately identify fungal samples at the species level. For a deeper overview of the underlying technology, see how MALDI-TOF mass spectrometry works for microbial identification
-
Why is MALDI-TOF more accurate than traditional phenotypic methods for filamentous fungi?
Unlike phenotypic methods that rely on visual morphology or growth characteristics, MALDI-TOF analyzes protein profiles, reducing human error and delivering consistent species-level accuracy. AccuFUN-ID® (as well as AccuPRO-ID® for bacteria and yeast) outperforms phenotypic methods and other MALDI-TOF systems based on extensive global comparison studies of environmental isolates from production facilities.
-
What types of filamentous fungi species can AccuFUN-ID® identify?
AccuFUN-ID supports identification of a wide range of filamentous fungi commonly found in pharmaceutical, medical device, and home and personal care environments. Our database includes over 17,300 bacterial and fungal entries representing more than 5,900 species for comprehensive coverage.
-
Is Accugenix® MALDI-TOF filamentous fungi identification compliant with regulatory standards (cGMP, ISO 17025)?
Yes. Our MALDI-TOF fungal identification process is CGMP-compliant and ISO 17025-accredited, meeting stringent regulatory requirements for pharmaceutical and medical device manufacturers.
-
What happens if my sample cannot be identified using MALDI-TOF?
If MALDI-TOF cannot identify your sample, we provide ITS2 DNA sequencing with AccuGENX-ID® at no additional cost. This maximizes the chance you receive an identification for every isolate submitted.
-
How do specialized Conidia IDFP improve fungal identification rates?
Conidia IDFP media promotes faster filamentous fungi growth and easier harvesting, enabling robust sample preparation for MALDI-TOF analysis. This specialized media significantly improves identification rates compared to standard industry practices.
